Background
SelectScience® has announced the first recipients of the new SelectScience® Seal of Quality program, recognising products that have consistently received the highest independent review ratings. The review process has been designed to assist scientists in making important purchasing decisions. The new Seals are assigned to approximately 0.1 % of products on the SelectScience® website and are allocated in line with strict criteria based on the quantity of reviews a product has received and the average rating of those reviews. Background / Problem
Common dose-response assays involve fixing different cell populations at a set number of time points to measure the response, for instance, at 3, 6, 12, and 24h of particle exposure. With DRAQ7™ we are able to measure the response every hour for 72h and because the measurements are taken from the same population of cells over time the results are not skewed by variations in cell population susceptibility to the drug. When used in combination with toxic QD we are able to achieve results which show particle dose over time and cellular response over time within the same system simultaneously.
Result / Solution
Our Cytometry group from the Engineering Department in Swansea University have relied on DRAQ7™ to perform live cell imaging in our work in collaboration with the Houston Methodist Research Institute, Houston. DRAQ7™ is easy to use, is non toxic and the signal remains stable for at least 72h (all our experiments ended at this time point). DRAQ7™ has allowed us to obtain several data sets which we hope to submit to high impact journals, one of which has already been accepted - Margulis-Goshen, K., et al., Active curcumin nanoparticles formed from a volatile microemulsion template. Needed a far-red fluorescent DNA stain that was broad spectrum in terms of cell types and worked with both live and fixed cells.

"CyGEL™ - suitable for complete immobilisation of LIVE protozoan parasites"

Background / Problem
The protozoan parasites Leishmania major and Trypanosoma brucei are causative agents of leishmaniasis and African trypanosomiasis, respectively. Live imaging of these organisms is hampered by the rapid flagellum-driven motility of these species.
Result / Solution
We found that CyGEL™ was suitable for complete immobilisation of procyclic stage L. major and T. brucei for 90 minutes without affecting cell viability. This has enabled us to analyse a GFP-tagged form of the L. major membrane-associated protein, HASPB, using live confocal imaging and FRAP.

DRAQ5™ is a valuable live cell reagent that is easy to use. It is our preferred live cell nuclear stain because the low intensity, low energy red excitation light it is gentle on living samples.
Preliminary data in our lab also showed improved cell health and higher migration speeds when comparing DRAQ5™ versus Hoechst as a live cell nuclear dye.
