Comments: DRAQ5 is fine unless you want to study DNA replication, repair, transcription, etc in living cells. Why? Because, like other DNA intercalators, DRAQ5 perturbs the DNA macromolecule and will evict histones and a variety of DNA binding proteins.

It is indeed a well-understood phenomenon that all cell-permeant DNA-binding probes will have some impact on cell behaviour. Accordingly, we alert users to this on the front of the product’s technical datasheet. Nonetheless, DRAQ5 has been widely used for live-cell end-point assays and as a means of identifying the nucleated cells in flow cytometry experiments on complex sample matrices such as bone marrow, besides its popularity as a standard IF counterstain.
