DRAQ5™ is the unique way to extract more power from the poor forgotten BD FACScan or BeckmanCoulter EPICS sitting in the corner of the lab!
Using either of these instruments with the far-red, live cell DNA dye DRAQ5™ you can simultaneously acquire FITC and PE data and get the cell cycle position of each population.
DRAQ5™ is excited as low as 488 nm yet its emission signal starts at 665 nm. That means NO COMPENSATION for the DRAQ5™ signal and three well-defined parameters.
In practice, nucleated cells are gated on DRAQ5™ in FL3 (FL4 on the EPICS for 4 parameter analysis - FITC, PE & ECD! See Plander et al, Cytometry 2003 for the evidence). This is great if you want to avoid the problems of antigen loss from lytic agents or want the speed of working with whole blood or bone marrow aspirates - non-nucleated cells are negative for DRAQ5™. FL1 and FL2 are then selected. Back-gating on the DRAQ5™ signal then gives cell cycle or ploidy information for each of these populations. If you do prepare MNCs or perform RBC lysis then you get full FSC and SSC information too. (See the Flow Cytometry pages for examples).
DRAQ5™ is so flexible that you can use it on LIVE or FIXED cells. It works in bone marrow, whole blood, MNCs and cultured cells. We supply DRAQ5™ ready to use. Store it in the fridge and there's no need for DMSO, RNase, .....
