The graph below shows standard fluorescence emission spectra for DRAQ5 in aqueous solution for a range of different excitation wavelengths. As can be seen, DRAQ5 can be effectively excited by 488nm systems. In these circumstances, we recommend that a long band pass filter be used - to capture as many emitted photons as possible.

 

DRAQ5 Em intensity at different Ex wavelengths

Excitation:

  • 647 nm line optimal (Exλmax 646 nm)
  • 488, 514, 568 and 633 nm lines, sub-optimal
  • Two-photon excitation (1047 nm) and excitation dark (700-850 nm)

Emission:

  • >665 nm to infra-red >800 nm (Emλmax 681 nm / 697 nm intercalated with dsDNA)
  • Minimal overlap with vis range e.g. GFP and FITC
  • Em. filters may include 695LP, 715LP or 780 LP

Multi-wavelength imaging with UV / vis fluorochromes

  • No fluorescence enhancement upon DNA binding
  • No photobleaching effect
  • Compatible with optics of benchtop flow, laser scanning cytometers and non-UV laser scanning and lamp-based confocal microscopes
