CyTRAK Orange™ spectral properties
A novel orange fluorescing nuclear/cytoplasm dye for live cells ....
CyTRAK Orange™ is excited in the low vis. range (457, 488, & 549 nm) with a profile similar to Propidium Iodide
- It can be used with a wide range of platforms: lamp/camera systems, HCS platforms, confocal microscopes and most commercially available flow cytometers
- There is no need to use UV to find cells (just like DRAQ5!) and the UV line is opened up for blue emitting labels such as CFP and Pacific Blue
- Harmful, high energy UV irradiation can be avoided where necessary
- Vis. range excitation avoids "white out" by UV-excited library compounds (commonly 10-15%) in screening assays
- CyTRAK Orange™ is co-excited with FITC/Cy2/eGFP and PE-Cy7: this easily allows two tagged targets plus the nucleus and cytoplasm with their respective borders
CyTRAK Orange™ is NOT excited by UV or red lasers (e.g. 633 & 647 nm)
- Further targets can be labelled in the UV channel (e.g. CFP) or the "red" channel (e.g. APC or far-red emitting mitochondrion labels) (see excitation spectrum)
CyTRAK Orange™ emits in the orange region with a peak at 615 nm
- No significant spectral overlap with co-excited green fluors (eGFP, FITC, Cy2): cell-based assays can be imaged in a single scan for the highest throughput
- Differential staining of nucleus and cytoplasm allows them to be seen as separate compartments to follow GFP-tagged temporospatial changes in live cells in a single acquisition from a 488 nm excitation
- CyTRAK Orange™ has an emission which can be separated from FRET dyes such as red-excited far-red APC-Cy7 and 488 nm-excited PE-Cy7 (see emission spectrum)
- CyTRAK orange™ has no appreciable autofluorescence minimising background
