Flow Cytometry residual leukocyte detection

Residual leukocytes are a common problem for platelet and red blood cell (RBC) preparations.
To detect residual leukocytes DRAQ5™ can be added to any remaining sample of platelets / RBCs and analysed by flow cytometry. Since platelets / RBCs contain no nuclear DNA, the DRAQ5™ labelling will clearly mark the nucleated leukocytes.
A final concentration of 5µM DRAQ5™ should be added to RBCs at 108 - 109 / ml.
Leukocytes (and nucleated erythroid and platelet precursors) will be detected by fluorescence signal above 665 nm preferably with long pass to increase sensitivity.
NB: This can be done retrospectively on any remaining sample up to several hours old.
